The assay with Azo-Carob galactomannan does employ 2% w/v Azo-Carob galactomannan. However, to generate the standard curve, the enzyme is first standardised using the Nelson/Somogyi reducing sugar assay using 0.2-0.5% carob galactomannan as substrate.
Why is Azo-Carob galactomannan (S-ACGLM) is prepared at 2% w/v for the assay, but carob galactomannan is used at only 0.2% w/v for the generation of the standard curve? Which concentration should I use?
