One unit is the amount of enzyme required to release one micromole of glucose reducing sugar equivalents (from oyster glycogen) per minute under standard assay conditions (10 mg/mL oyster glycogen; pH 3.5; 40˚C). Reducing sugar is measured with the Nelson/Somogyi Reducing Sugar method.
