The glucose measurement (incubation with GOPOD Reagent) is linear between 4 and 100 micrograms of glucose per assay, which is an equivalent of 0.35 to 8.5% of β-glucan in the powder samples, when the streamlined method is followed as described in the data booklet. The absorbance received for your sample must not exceed the absorbance values obtained for 100 μg of glucose standard (0.1 mL of standard at 1.0 mg/mL that is supplied with the kit). If an expected β-glucan content in your sample is higher than 8.5% or received absorbance values for the sample are higher than the absorbance values obtained for the glucose standard, the sample solution must be diluted sufficiently with 200 mM sodium acetate buffer (pH 4.0) before the β-glucosidase treatment. For example if a sample contains 20% beta-glucan it should be diluted 3-fold with 200 mM sodium acetate buffer (pH 4.0) before dispensing aliquots for incubation with β-glucosidase.
