Corn starch does not contain beta-glucan (unless added). The K-YBGL method was designed for measurement of beta-glucan in mushroom where beta-glucan is high and alpha-glucan is low. In cases where beta-glucan is low and alpha-glucan is high, you are subtracting one high absorbance value from another. The errors in both methods are +/- 2% (at best). The K-YBGL assay kit can easily show which samples are loaded with alpha-glucan, but accuracy of beta-glucan content is not great, due to the above limitation.
