At Megazyme, the K-TDFR method has been validated with the use of glass beakers in a shaking water bath.
The assay can be adapted the assay to suit different laboratory needs, however it would be up to the customer to validate this change.
Some key things to note:
- Ensure that the correct temperature can be reached with your equipment.
- Ensure sufficient shaking of the sample so there are no clumps. This can effect enzyme accessibility to the whole sample.
- Check the temperature of your samples for the alpha amylase incubation as heat transfer is faster in glass than plastic.
- You would need to ensure that the temperature of your sample within the new equipment reaches the desired temperature of 98-100 degrees for the alpha amylase incubation. (>95 degrees is sufficient.)
