It is possible to increase the amount of sample used in this assay, we have not validated this in our labs so customers will need to validate this internally.


If you wish to do so and subsequently need to increase the amount of ethanol added, you must take this into account for the calculations.


The sample does not need to be completely dissolved by the ethanol however it does need to be completely wetted by the ethanol, this is important as this aids the dissolution when sodium hydroxide is added.


The volumes of all other components should remain the same.

If you do alter the amount of ethanol please see example below of final extraction volumes. This new extraction volume will be required for calculations, this can be easily adjusted in the MegaCalc calculator: Total Starch (K-TSTA) Mega-Calc

For example:


for 100mg, 0.2mL of 80% ethanol is added, Extraction Volume = 10.4 (no changes to the procedure)
for 200mg, 0.4mL of 80% ethanol is added, Extraction Volume = 10.6
for 300mg, 0.6mL of 80% ethanol is added, Extraction Volume = 10.8 

To ensure the assay in running correctly we recommend run the kit control through this procedure and if they correct result is obtained you can conclude the assay is working correctly.
