Are there any tolerances on the temperatures stated in the Total Starch Assay Kit (K-TSTA) Procedure?

We recommend a tolerance of 50 +/-0.5˚C for the enzymic digestion. 


The colour development temperature is a bit more flexible and can be done at 40˚C or 50˚C. This is an end-point assay. 

The reaction is complete after approximately 15 min at 40˚C, however we recommend incubating the samples for 20 min to ensure that the reaction has gone to completion. The main thing is to run samples and glucose standards with GOPOD Reagent at the same time in a water bath.
