It is necessary to filter because of the haze associated with the flour slurry. If you centrifuge all of the haze will not be removed and this gives the higher absorbance values.
In Amylazyme tablet (T-AMZ) assay procedure can centrifuging be used in the final clarification step rather than filtering? The results differ by 0.5 absorbance units.
