The acid hydrolysis procedure of K-YBGL gives complete hydrolysis of all glucan, α- and β- as well as all mannan (which is α-linked). K-MANGL allows measurement of total glucose and total mannose; so total glucan and total mannan. In sample preparation examples in K-MANGL booklet (“Determination of D-glucose and D-mannose in polysaccharides”) we recommend using ethanol washing step to remove low degree of polymerisation (DP) sugars, followed by acid hydrolysis (as in K-YBGL) prior to the enzymic determination of free glucose and mannose.
