Is there an appropriate stopping point to run the K-YBGL assay over two days?

With the recent update of the K-YBGL assay kit (September 2025), the overall assay time has been significantly reduced from 7 hours to 4 hours and 25 minutes, facilitating the integration of the procedure into a single day.

 

Storage of hydrolysates overnight at 4°C prior to performing the GOPOD (Glucose Oxidase/Peroxidase) assay was evaluated in order to determine the feasibility of incorporating a pause point in the Total and α-Glucan analytical workflows without compromising data integrity. The findings indicate that α-Glucan measurements remain stable after overnight storage of hydrolysates (from Step 6 in Method B) at 4°C. However, Total Glucan results show a statistically significant shift when hydrolysates are stored overnight before testing (from Step 8 in Method A). Consequently, introducing a pause point is method-dependent: it is acceptable for α-Glucan analysis but not recommended for Total Glucan without further validation or adjustment.
