Do I need to have two sets of glucose standards and reagent blanks for K-YBGL?

It depends on how you run your assays.


If you measure glucose from both the total glucan and α-glucan procedures within the same batch, one set of reagent blanks and one set of quadruplicate glucose standards is sufficient. In Megacalc, simply enter the same values in both sections.


However, if you measure glucose from the total glucan procedure separately from the glucose generated in the α-glucan procedure—i.e., in two separate batches—then you’ll need two separate sets of glucose standards and reagent blanks: one for each batch.


Megacalc should be completed with the appropriate data for each set.
