Information for analysing MUVA-JO-1437 and MUVA-JO-1438 with K-DLATE D/L-Lactic acid kit.

The same sample preparation can be used for both samples, for Neogen Lactic acid kits.

 

  • Weigh ~ 5 g of yoghurt sample into a 50 mL volumetric. 
  • Add ~ 30 mL of distilled water. 
  • Add 5 mL diluted Carrez I and mix thoroughly. 
  • Add 5 mL diluted Carrez II and mix thoroughly. 
  • Add 1 mL 1 M NaOH, and mix thoroughly. 
  • Prepare to the final volume of 50 mL with distilled water in the volumetric flask. 
  • Filter with Whatman No.1 filter paper or equivalent. 


Use the clear filtrate, after subsequent dilution if necessary as the sample for the assay.


This preparation is for use with the K-DATELQ / K-LATELQ or the K-DLATE assay kits.

 

Dilutions required after preparation:

 

For K-DLATE individual D and L lactic acid tests should be performed. 

Both samples are prepared as above. For D-lactic measurement with the K-DLATE kit, no further dilution is required after preparation, use 0.1 mL of the clear filtrate in the assay.

For L-Lactic acid measurement with the K-DLATE kit, a 10-fold dilution is required after preparation. Use 0.1 mL of the 10-fold dilution in the assay.


For K-DATELQ (D-Lactic acid) no further dilution is required for either sample. Use 0.1 mL of the filtrate in the assay.

For K-LATELQ (L-Lactic acid) a 10 fold dilution is required. Use 0.1 mL of the 10-fold dilution in the assay.

 

Total Lactic acid content

 

Total Lactic acid = D-Lactic Acid content PLUS L-Lactic acid content.
