Protein Digestibility Assay Kit (K-PDCAAS)

Why does the calculation for the K-PDCAAS kit only use the amino acids, Proline, Lysine, Histidine and Arginine?
All amino acids are taken into account however Proline, Lysine , Histidine and Arginine have to be adjusted for their ninhydrin reaction properties.  All o...

Why is the molecular weight of water subtracted for the calculation of amino acids for the K-PDCAAS assay kit?
"Concentrations of amino acids in the sample are calculated using the molecular weight of each individual amino acid minus the molecular weight of wate...

How is it possible to obtain values above 1 for in vitro digestibility and amino acid score using the K-PDCAAS calculation?
Values above 1 are possible for digestibility.  It means that they are higher than the digestibility of casein which is given a value of 1.  Results above 1...

Do the controls and casein need to be tested with ever batch of samples for the Protein Digestibility Assay Kit (K-PDCAAS)?
For every batch of new samples that are tested using the Protein Digestibility Assay kit (K-PDCAAS), it is recommended that all standards plus casein a...

How long is (K-PDCAAS) glycine calibration curve valid for?
The method states the stock standard is good for 3 months and the glycine standard curve is good for 1 month.   A new calibration curve should be performed...

Can insoluble proteins like plant proteins be analysed (around 60-70 % soluble at pH above pI) using the K-PDCAAS Assay kit?
The assay is run at pH 2.0.  Any proteins soluble at this pH will not be digested or included in the measurement. The protease enzymes will not interact...

Will protein insolubility (turbidity/suspension) affect absorbance measurement for the PDCAAS Assay?
Yes it will interfere with the absorbance measurement.  There is a TCA precipitation step and a centrifugation step which should clarify the sample.

Can a low digestibility result due to low solubility at neutral pH be inferred as low PDCAAS?
As the method is run at pH 2 insolubility at neutral pH will not be involved.

Which protein is used as standard or control protein for the PDCAAS Assay?
As per the procedure in described in the kit booklet Casein and several other undisclosed standards are used as reference of this method to the in-vivo metho...
